Characterization of CD4+ and CD8+ T cell Responses in HIV-1 Clade C Infection

Danni Ramduth, Thumbi Ndungu

This study proposes to characterize the immunogenicity of HIV-1 specific CD4+ T cells in chronically infected therapy naive adults and acutely infected infants (one arm receiving continuous antiretroviral therapy and one arm therapy na´ve). áSpecifically we will assess HIV antigen specific secretion of interferon gamma; interleukin-2 and tumour necrosis factor alpha. We will also investigate proliferative activity and the association between HIV-1 specific CD4+ and CD8+ T cell activity and their associations with viral loads. These aims will be accomplished by the following techniques: the Elispot assay, Intracellular cytokine staining, Proliferation assays and measurement of CD4 counts and viral loads.
