The extent of pks12 variation in clinical strains of Mycobacterium tuberculosis circulating in KwaZulu-Natal and Maputo

Abstract


Aim of study: To determine the variation in the pks12 gene amongst clinical isolates of Mycobacterium tuberculosis circulating in KwaZulu-Natal, South Africa and Maputo, Mozambique.

Specific objectives:

a.     To type isolates obtained from Tugela Ferry in KwaZulu-Natal and Maputo (Mozambique) by means of spoligotyping and restriction fragment length polymorphisms (RFLPs)

b.     To select isolates from each area based on the typing pattern obtained

c.     To amplify and isolate pks12 gene from the selected isolates using long range PCR

d.     To use DNA sequencing to examine the variation in pks12 amongst isolates from different clusters and amongst isolates from the same RFLP-spoligotype cluster (intracluster comparison).
